Protein-protein interactions: co-evolution, prediction and design

Résumé :
In order to analyze protein-protein interactions, we focus on the combined use of structural data and evolutionary information.
In a study of over 1,000 couples of homologous heteromeric interfaces, we uncover astonishing plasticity in the way interface structure evolves, although we identify some rather invariant features which provide tracks for extracting meaningful information from multiple sequence alignments of binding partners.
Consequently, we develop a knowledge-based potential relying on evolutionary interface descriptors to improve the prediction of protein interfaces.
Finally, a better understanding of protein-protein interactions has major implications with regard to the design of specific interaction inhibitors.
